  • The project entitled “Ex-vivo modeling of lymph node microenvironment and its impact on drug response in lymphoma” was funded by the DFG from 01.12.2021 to 01.12.2024 and was focused on establishing an advanced 3D co-culture system that accurately replicates the architecture and ecosystem of follicular lymphoma (FL) in order to study the ex vivo drug response. Throughout the project, Prof. Dietrich’s group successfully developed a multiplex immunofluorescence-based detection method. Using the 56-plex co-detection by indexing (CODEX) antibody panel, they compared the tumor architecture and microenvironment between non-malignant reactive lymph nodes (rLNs), diffuse large B-cell lymphoma (DLBCL-LNs), and FL-LNs. They identified 21 distinct hematopoietic cell types as well as non-hematopoietic structure-defining cell types or cell states. Their study revealed that cellular neighborhoods were conserved mainly between nonmalignant and FL-LNs but were altered due to the malignant expansion of FL cells. Additionally, their results demonstrated significant changes in stromal cells, underscoring the pivotal role of stromal cells in FL tumorigenesis.
